Overview

Well-versed practitioners detail powerful affinity chromatography methods, ranging from such traditional affinity purification as immunoaffinity chromatography, to the use of state-of-the-art phage-display technology in the discovery of affinity ligands and drugs. Also included are effective and reproducible separations of such small molecules as haptens, protein ligands, and supramolecular structures. Each chapter is devoted to a specific technique and includes an introduction, an explanation of principles, a detailed materials list, and step-by-step instructions that ensure experimental success. Practical notes suggest alternative procedures and describe how to overcome problems that may occur. State-of-the-art and eminently instructive, Affinity Chromatography: Methods and Protocols offers novice and experienced scientists alike a comprehensive collection of proven and readily reproducible techniques that are ideally suited for use not only in drug discovery and development, but also in work on affinity interactions, fermentations, separations, drug targeting, and cell culture.

This book is a useful guide for new and experienced practitioners in the field of affinity chromatography. J. Am. Chem. Soc. The principles are first described, followed by a list of materials, and the method to be used. Probably the most useful section is that entitled Notes', which defines experimental parameters and problems that may occur. for this reason the much larger audience of experienced users may be attracted to this book if only to reduce the inordinate amount of time often spent in optimizing a specific separation. The 19 chapters of the book cover the whole range of available affinity techniques in a most concise manner. Just sufficient theory is offered to provide an understanding of the principles, and includes the most recent applications with two chapters on phage display. It is adequately indexed, concise, readable and virtually free from typographical errors. - Chromatographia the authors describe very different aspects of affinity chromatography; many parts of this book are pleasant reading. For all biochemical libraries it is simply a must. - Chembiochem The title clearly describes the scope - and the reader gets what he/she expects. the authors have compiled information in very helpful tables, which would be difficult to obtain from other sources, for instance, the binding and elution conditions for different lgG subclasses (human/mouse/rat) on Protein A. For all biochemical libraries it is simply a must. - Chembiochem this volume should be a useful handbook in any laboratory that is using, or planning to use, affinity chromatography techniques. - Biomedical Chromatography This handbook has the ambitious purpose of giving a practical overview of a field which concerns almost any researcher in biology nowadays: that of affinity chromatography. It really answers a need, as chromatographic steps are too often overlooked in the Materials and methods' sections of scientific publications. Here, protocols are precisely described with plenty of troubleshooting advice. Although the book is mostly intended for new partitioners (both industrial and academic), it will certainly also provide experts with valuable information, as the field of affinity chromatography has becomes fantastically diverse since the concept was first introduced in 1968. The focus chosen by the editors is very down-to-earth: this approach will no doubt be useful to those who seek to master technical aspects the publication of this book is an excellent and timely initiative. - Biochimie Humana's Methods in Molecular Biology series remains an excellent candidate for standing order status in any university science or medical library. This reviewer has been impressed how many new and experienced scientists like the various titles, and how many volumes have been ordered by individuals because they are so necessary for lab bench use, a four week loan is just not enough. With excellent worldwide contributors, clear and concise methods, the always-useful Notes section for each protocol, and the generally extensive and up-do-date reference lists, these books are good additions to the Humana series. Like Methods in Enzymology, Methods in Molecular Biology is indexed in PubMed, making it very easy to find the protocols within. - E-Streams Overall this text provides an excellent review of the latest trends in separating and quantifying specific molecules such as peptides and DNA and should prove to be a required text for in the age of biotechnology. - Die Pharmazie This book fills a worthwhile niche with chapters covering major applications explicitly pertinent to cutting-edge life science research...This book stands out from the pack in that it addresses practical affinity application areas that have potentially broad use in modern technology. - Journal of medicinal Chemistry This book is an excellent up-to-date collection of carefully selected methods for affinity chromatography of small molecules, such as haptans, to supramolecular structures, such as in phage display. This book would be a most useful guide for those scientists involved in the separation and analysis of macromolecules, such as biochemists, biologists and chemists, and health scientists. - Cell Biology International It is an excellent book, and can be recommended for every laboratory involved in the synthesis and analysis of chiral compounds. Moreover, the editors have succeeded in getting internationally known leading experts to write the contributions. - Angewandte Chemie
